Abstract

Thiol-dependent peroxidases, as peroxiredoxins, play a role in the response of oxidative stress due to reactive oxygen species (ROS) accumulation. There are five isoforms of peroxiredoxins in Saccharomyces cerevisiae, localized in different cellular compartments. Prx1p is a mitochondrial isoform and plays an important role in the elimination of hydrogen peroxide produced during the electrons transport chain in mitochondria. As mitochondria is the major intracellular ROS producer, much interest has arising regarding Prx1p. Thereat, this project aims to expand the knowledge about this enzyme, seeking to determine its localization in differents submitochondrial compartments, as well as its involvement in mitochondrial redox processes regulation. Recent studies have shown that mitochondrial ROS act directly in autophagy regulation, a highly conserved process involved in cellular components recycling. This process has gained prominence as indicator of proteins and organelles quality. As Prx1p acts in mitochondrial ROS metabolism, we seek to evaluate its possible role in autophagic response regulation. Since there is also a mitochondrial peroxiredoxin system in mammals, this study may have relevance in the understanding of autophagy regulation in these organisms.
