Bone remodeling and biomechanical analysis of maxillary alveolar bone in rats with masticatory overload

Abstract

Studies have used different methodologies for stress evaluation in bone tissue, mainly in the alveolar bone, with presence of oclusal changes, once the loads applied on the teeth are dissipated to alveolar bone. Objective: To evaluate the bone remodeling and alveolar bone biomechanics in adult rats with occlusal stress by premature contact. Materials and methods: Fifth adult (2 months old) and male rats (Rattus norvegicus albinus), Wistar lineage will be used. After completed 2 months old, 40 animals (experimental group) will be subjected to unilateral (right side) cementation of a metallic wire on the occlusal surface of first maxillary molar, featuring a premature contact condition. Then, these animals will be distributed in 4 subgroups (n=10) according to the sacrifice periods: 7, 14, 21 and 28 days after cementation. In control group, the teeth will be kept without occlusal changes. After initial period (animals with 2 months old), these animals (n=10) will be sacrificed after 28 days. The head of all animals will be subjected to micro-computed tomography scanning, using a SkyScan 1174 micro-tomography. After the three-dimensional reconstruction of both sides of maxillary bone, the volume of alveolar crest will be performed using the CTAnalyzer software. The three-dimensional models will be also used to simulate the masticatory function with occlusal changes in finite element analysis. The stress dissipation will be evaluated in the alveolar bone crest. The pieces directed to histologic and immunohistochemistry evaluation will be subjected to histological process involving demineralization, dehydration, diaphanization, paraffin inclusion and microtomy. The histologic slices will be stained by hematoxylin and eosin to evaluate the feature of citoarchitecture. The slices will be also used to immunohistochemistry process to evaluate the proteins expression related to extracellular bone matrix: osteopontin and osteocalcin, which feature the bone tissue mineralization, Type I collagen, main protein of extracellular bone matrix, TRAP, for osteclast determination, OPG and RANKL, to characterize the responses in bone remodeling. EXPECTED RESULTS: The association between the results in experimental and computation simulation, which were proposed in this study, will provide the better understanding of the relationship between these occlusal changes and the mechanobiology responses in alveolar bone tissue. Thus, this knowledge will be applied to studies with clinical application in Dentistry. (AU)
